Role: Upstream Principal Scientist, Vector Biology, focusing on early process development and establishing manufacturable lentiviral vector platforms for in-vivo CAR-T programs. Responsibilities: optimize seed train and bioreactor workflows from shake flask to 40L scale; evaluate and engineer LVV constructs, including plasmid design and vector optimization; develop and refine upstream processes such as transfection and stable cell line production; collaborate cross-functionally with downstream, analytical, and technical teams to ensure process coherence; generate data packages for process lock and regulatory filings; author technical reports and communicate findings to leadership. Requirements: MS with 10+ years or PhD with 5–8+ years in bioengineering, cell biology, or related fields; proven hands-on experience with viral vector upstream process development, mammalian suspension culture, and bioreactor operation; expertise in molecular biology techniques relevant to LVV construct design; familiarity with downstream processing and integrated platform development; strong data analysis, reproducibility, and scientific communication skills. High-Value: focus on early-stage process optimization for lentiviral vectors used in CAR-T therapies; experience with scalable bioreactor processes (up to 50L); GxP and biosafety compliance; collaborative work environment in R&D; based in Philadelphia, PA.
