Role Summary
We are seeking a curious and motivated Senior Scientist to join our Regeneron Genetic Medicines group, working on oligonucleotide-based therapeutics. You will be spearheading assay development, candidate screening, validation, and structure-activity relationship (SAR) optimization. The role involves siRNA design, high-throughput screening (HTS), off-target analysis, and molecule optimization to support therapeutic development.
Responsibilities
- Design and optimize siRNAs for target specificity and efficacy.
- Lead high-throughput screening (HTS) and secondary screening, including dose-response studies to determine IC50.
- Develop and validate screening assays (e.g., bDNA, RT-qPCR).
- Implement automation for HTS and optimize screening systems (e.g., primary cells, reporter systems).
- Establish off-target screening approaches.
- Use off-target data to guide SAR optimization and improve molecular design.
- Lead a multidisciplinary team, ensuring alignment with project goals.
- Collaborate with bioinformatics, automation, and chemistry teams.
Qualifications
- Required: Ph.D. in Molecular Biology, Biochemistry, or related field.
- Required: 3+ years of experience in RNAi research, high throughput screening, and/or SAR optimization.
- Required: Expertise in siRNA design, assay development, and off-target analysis.
- Required: Proficiency in RT-qPCR, including probe designs and multiplex assay development.
- Required: Experience with primary cells and reporter systems.
- Required: Experience with automation systems and bDNA assays to support HTS.
- Required: Knowledge of computational tools for transcriptome analysis.
- Required: Background in drug discovery workflows.
- Preferred: Strong attention to detail, rigorous scientific approach, and effective communication across all levels.
