Position Overview
Senior/Principal Scientist in ADC Conjugation Process Development to optimize critical conjugation and purification process parameters for ADC Drug Substance (DS) development, coordinating work from milligram to multi-gram scales (including tox/GMP batches).
Responsibilities
- Implement ADC Conjugation Process Development strategies using DOE and OFAT for critical reaction parameters.
- Scale ADC batches from milligram to >100 g for PD optimization, analytical/formulation development, DS/DP stability studies, and in vivo experiments (including GLP tox) for IND and BLA activities.
- Lead multi-gram scale purification optimization for antibody intermediates and ADCs via UF/DF, and, if needed, preparative chromatography or other approaches.
- Serve as ADC DS SME for internal CMC teams and external CDMO partners; occasional travel to advise/oversee Development and GMP manufacturing.
- Prepare SOPs, protocols, and reports for tech transfers, GMP activities, and regulatory submissions.
- Stay current with ADC DS trends/innovations/regulatory requirements and integrate beneficial updates into internal strategies.
- Lead/support authoring and review of Module 3 regulatory sections (IND/IMPD/BLA) and respond to information requests with global health authorities.
Qualifications
- BS/MS/PhD in Chemistry, Chemical/Biochemical Engineering, Biotechnology, or related field.
- Minimum experience: 5+ years (PhD), 8+ years (MS), or 10+ years (BS); ADC or Conjugation/Bioconjugation Process Development focus required.
- Demonstrated ADC conjugation technology/strategy development and scale-up with experience across linker-payloads and antibody intermediates.
- Prior tech transfer and external CDMO engagement (Development and GMP) plus cross-functional coordination.
Knowledge and Skills
- Strong grasp of ADC conjugation, purification, analytical, and characterization workflows.
- Multi-gram TFF expertise for removal of unreacted linker-payloads and stable ADC DS formulation.
- Analytical methods: SEC, HIC, RP/U-HPLC; LC-MS, icIEF, CE-SDS.
- GMP standards/regulatory knowledge supporting IND, INDa, and BLA submissions.
- Ability to manage multiple projects and balance hands-on vs. strategic responsibilities.
- Exceptional collaboration and communication.
