Key Responsibilities:
- Express novel multi-specific biologics formats to support therapeutic area drug discovery.
- Generate and screen stable CHO cell lines with optimized attributes (titer, monomer, chain-pairing) using 2+ vectors.
- Determine optimal chain ratios for 3- and 4-chain molecules in CHO cells using multiple selection strategies and process optimization.
- Perform process development/optimization for batch and fed-batch bioreactor and shake flask expression.
- Characterize stable CHO line copy number and mRNA transcript levels using ddPCR and related tools.
- Apply targeted integration technologies, including FACS-based single-cell sorting to generate mini pools.
- Leverage automation and high-throughput workflows to increase efficiency.
- Support data digitization (electronic data capture, analysis, reporting).
- Troubleshoot expression, stability, glycosylation, aggregation, and chain pairing issues.
- Purify and conduct analytical biophysical characterization (analytical HPLC for titer, monomer, chain pairing).
- Independently design, execute, and interpret experiments; analyze, document, and report results.
- Handle stable CHO pools/cell lines (thaw, culture, expression, cryopreservation).
Qualifications:
- BS in biology/chemistry/biochemistry (typically 5+ years) or MS (typically 2+ years).
- Hands-on experience with CHO expression of monoclonal antibodies, bi-specifics, and multi-specifics (thaw, culture, expression, cryopreservation).
- Prior stable CHO cell line generation for complex multi-specifics.
- Prior process development/optimization and automation experience.
- Desired: analytical characterization of multi-specifics; targeted integration, FACS, ddPCR; vector design proficiency.
- Desired: mammalian expression using Wave bioreactors.
- Desired: demonstrated ability to resolve challenges (poor chain pairing, low expression, aggregation, glycosylation).
