About Stoke:

Stoke’s initial focus for its TANGO platform is on diseases of the central nervous system and the eye that are caused by a loss of ~50% of normal protein levels (haploinsufficiency).

Stoke has identified STK-002 as a clinical candidate for the potential treatment of autosomal dominant optic atrophy (ADOA), the most common inherited optic nerve disorder. The company is also pursuing a potential new medicine for Syngap1, a severe and rare neurodevelopmental disorder, in collaboration with Acadia Pharmaceuticals.

Proof of concept has been demonstrated in other organs, tissues, and systems, supporting broad potential for the Company’s proprietary approach.

Position Purpose:

We are seeking a highly motivated and detail-oriented Formulation Scientist to join our Early Development team focused on oligonucleotide-based therapeutics. This role offers a unique opportunity to contribute to the design and development of innovative drug formulations at the preclinical and early clinical stages. You’ll work closely with cross-functional teams to support formulation screening, in vitro and in vivo formulation preparation, and process development for novel oligonucleotide candidates. This position will report into Senior Principal Scientist, Formulation Lead.

Key Responsibilities:

  • Design and execute formulation studies for oligonucleotide drug products
  • Prepare prototype formulations and conduct pre-formulation characterization (e.g., solubility, pH stability, buffer selection)
  • Collaborate with analytical, process development, and manufacturing teams to ensure seamless tech transfer
  • Support analytical testing and interpretation of data related to physical and chemical stability
  • Maintain accurate and timely documentation of experiments, protocols, and results in electronic lab notebooks
  • Contribute to the preparation of technical reports, presentations, and regulatory documentation
  • Stay current with scientific literature and industry trends in oligonucleotide formulation and delivery
  • Additional duties as assigned

Required Skills & Experience:

  • PhD in Pharmaceutical Sciences, Chemistry, Biochemistry, or related field, Master’s degree with 6+ years of experience in formulation development, or Bachelor’s with 10+ years of experience
  • Experience with nucleic acid-based therapeutics preferred
  • Familiarity with oligonucleotide properties, degradation pathways, and formulation challenges
  • Hands-on experience with lab techniques such as pH adjustment, UV spectroscopy, endotoxin and physical property measurement; hand-on experience with HPLC/UPLC is a plus
  • Strong organizational skills and attention to detail
  • Excellent communication and teamwork abilities

Stoke operates in Bedford, MA. This position is a lab-based position in our Bedford, MA location.

Travel:

This position will require approximately 5% travel.
