Role Overview:
Legend Biotech seeks an experienced Scientist (Upstream team, Vector Biology) in Philadelphia, PA focused on in-vivo CAR-T programs. Own hands-on execution and data-driven development of lentiviral vector upstream processes, including seed train strategy and scale-up from small-scale to 40-L bioreactor runs. Partner cross-functionally to support end-to-end process alignment, comparability strategy/readiness, and efficient technology transfer.

Key Responsibilities:
- Define and optimize upstream process from shake flasks through bioreactors; scale to 40-L with reproducible performance.
- Execute and optimize lentiviral vector production workflows for in-vivo CAR-T, including transient transfection and/or stable cell line optimization.
- Conduct media optimization, feeding strategy development, and process control adjustments to improve yield, quality, scalability, and robustness.
- Develop and execute fit-for-purpose scalability/readiness studies.
- Support technology transfer to Technical Development; contribute to experimental design and comparability discussions.
- Plan, perform, analyze, and troubleshoot experiments with minimal supervision.
- Deliver data summaries, recommendations, and high-quality technical reports/slide decks.
- Adhere to BSL-2 standards; maintain compliant ELN documentation and ensure data integrity/reproducibility.

Requirements:
- MS with 4+ years; or PhD with 1 year prior industry experience in Bioengineering, Virology, Cell biology, Chemical Engineering, or related.
- Hands-on viral vector production and characterization.
- Mammalian suspension cell culture, transient transfection, and upstream bioprocess development.
- Molecular biology skills: plasmid construction; PCR/qPCR/dPCR assays; cell line development.
- Experience scaling from shake flasks to bioreactors (up to 50 L preferred).
- Gene therapy platform experience preferred (lentiviral vectors).
- Strong analytical/problem-solving, organization, communication, and ability to work independently and cross-functionally.

Pay Range (Base Pay): $96,267–$126,351 USD.
