Role Summary

This on-site role is located in Cambridge, MA, with no remote work or relocation assistance. Local candidates only will be considered. The Research Scientist I/II will join the xRNA Therapeutics team to perform hands-on oligonucleotide synthesis, purification, and analysis, supporting the growth of internal synthesis capabilities under the supervision of a Principal Scientist. The role requires a proactive, collaborative approach in a fast-paced, interdisciplinary environment.

Responsibilities

  • Execute solid-phase synthesis of research-scale oligonucleotides with various chemical modifications using automated DNA/RNA synthesizers such as Shasta, K&A, and Mermade.
  • Perform purification, desalting, and analysis of oligonucleotides using techniques including reverse-phase, ion-exchange HPLCs, UPLCs, and LCMS.
  • Method development and optimization for improved synthesis, purification and analysis of oligonucleotides and conjugates.
  • Prepare and maintain standard operating procedures (SOPs) for synthesis, processing, and purification of therapeutic oligonucleotides to ensure reproducibility.
  • Operate, troubleshoot, and maintain critical laboratory equipment such as synthesizers, HPLCs, and LCMS.
  • Organize experimental data and maintain a well-documented electronic notebook (eLN).
  • Demonstrate high motivation and productivity in a fast-paced environment.

Qualifications

  • Preferred: Research Scientist I level – at least 1 year of chemistry laboratory experience in academic or industrial settings.
  • Preferred: Research Scientist II level – at least 2 years of chemistry laboratory experience in academic or industrial settings.
  • Required: Familiarity with automated solid-phase synthesizers and therapeutic oligonucleotide synthesis and downstream processing.
  • Required: Proficiency in purifying, analyzing and characterizing oligonucleotides via LC/HRMS, RP-HPLC, AEX-HPLC, SEC-HPLC, and UV-vis analytics.
  • Required: Basic understanding of RNA therapeutics and their role in biology.
  • Required: Be a team player prepared to work in a collaborative, fast-paced environment with a high degree of flexibility and self-motivation.
