Role: Principal Scientist specializing in T cell receptor (TCR) optimization for next-generation TCR-based therapeutics at Moderna. Responsibilities: develop scalable workflows using display technologies and directed evolution to enhance affinity, specificity, stability, expression, and developability of TCR candidates; design rational libraries and lead iterative multi-parameter optimization campaigns; collaborate with computational teams on cross-reactivity and structural modeling; interpret complex biophysical and functional datasets; support TCR modalities including TCR-T and TCR engagers; evaluate new screening and engineering technologies; communicate findings to multidisciplinary teams. Requirements: PhD in immunology, protein engineering, biochemistry, or related field; 5+ years in TCR or protein engineering, display technologies, and functional assays; hands-on experience with affinity maturation, specificity engineering, and developability; proven ability to lead complex experimental workflows and collaborate across teams. Preferred: strong knowledge of TCR-pMHC interactions, cross-reactivity assessment, NGS data analysis, and experience with automation and new platform technologies. High-Value: biopharma therapeutic development in immune-oncology, TCR-mimetic modalities, active involvement in early to late-stage preclinical programs. WorkSetup: hybrid (onsite in U.S.), with collaboration across cross-functional teams.
