Position Overview
- Postdoctoral Scientist (DDCS) focused on CNS gene therapy biodistribution and molecular bioanalysis to optimize and advance viral and non-viral gene therapy platforms for neurological/neurodegenerative diseases.
Key Responsibilities
- Design, develop, optimize, and execute biodistribution assays for AAVs, LNPs, siRNA distribution, transgene expression, and persistence across CNS and peripheral tissues.
- Apply vector biology/AAV serotype-specific biodistribution expertise (e.g., AAV9, AAVrh10, PHP.eB).
- Use molecular/bioanalytical methods: PCR/qPCR/ddPCR, ELISA/immunoassays, LCβMS (as applicable), IHC and spatial tissue analyses, and NGS.
- Develop off-target analyses and DRG toxicology assays; ensure assays are fit-for-purpose (sensitive, reproducible, aligned with translational goals).
- Independently design/execute studies using DoE principles; troubleshoot assay/tissue/vector detection issues; maintain rigorous documentation.
- Integrate biodistribution with PK/PD to inform dosing, route, and therapeutic index; link molecular readouts to functional/anatomical/disease endpoints.
- Oversee CRO-led in vivo biodistribution studies and ensure data quality/timelines.
Basic Requirements
- PhD in Molecular Biology, Biochemistry, Pharmacology, Neuroscience, Bioengineering, or related field.
- In-depth AAV biodistribution analysis and/or molecular bioanalytical assay development.
- Hands-on PCR/qPCR/ddPCR, ELISA, IHC, NGS, and/or related techniques.
- Strong CNS gene therapy delivery/transgene expression/tissue biology knowledge; independent experimental design, troubleshooting, and documentation.
- US work authorization required; employer will not sponsor visas.
Additional Preferences
- PK/PD knowledge; spatial transcriptomics/RNAscope; CNS tissue/animal model experience; imaging/functional integration; CRO collaboration; familiarity with FDA long-term follow-up guidance and AAV safety guidance.
Other Information / Benefits
- Fixed-term: 1 year, extendable up to 4; daily on-site lab work in Boston, MA.
- Anticipated wage: $58,000β$123,200; eligibility for company bonus (FTE) and comprehensive benefits (e.g., 401(k), medical/dental/vision, time off).
