Clinical Research and Development
Main Responsibilities:
- Maintain/expand/quality-control human iPSC lines; generate/validate isogenic disease-relevant lines using CRISPR/Cas9, base editing, and prime editing (knock-in/knock-out/tagged reporters).
- Differentiate iPSCs into CNS cell types (microglia, astrocytes, oligodendrocyte-lineage cells, motor/cortical neurons); optimize co-culture/tri-culture systems.
- Build/validate fit-for-purpose cellular assays for TDP-43 biology and other mechanisms (cryptic exon readouts such as STMN2/UNC13A/HDGFL2; TDP-43 mislocalization/aggregation; stress responses; functional endpoints).
- Develop high-content imaging and plate-based assays (96/384-well) for screening, target validation, and MoA; partner to qualify assays internally.
- Apply AI/ML image analysis for cellular phenotyping.
- Design/execute/analyze pooled and arrayed CRISPR screens (CRISPRko/i/a), including lentiviral delivery, selection (FACS/imaging), and single-nuclei RNA/ATAC-seq sample prep.
- Generate/maintain stable cell lines, lentiviral reporter systems, and inducible expression systems.
- Maintain electronic lab notebooks, present results, and contribute to publications/posters/patent filings.
- Develop/optimize/document SOPs; train junior team members.
- Coordinate with academic collaborators and CROs on cell line generation, screen execution, and assay transfer.

Education/Required Qualifications:
- BS with 4+ years OR MS with 2+ years in stem cell biology/neuroscience/molecular/cell biology (or related).
- Hands-on human iPSC culture, banking, QC (karyotype/STR/pluripotency), and directed differentiation into at least one CNS cell type.
- Hands-on plate-based and/or high-content imaging assay development (miniaturization, Z’-factor/reproducibility, basic stats).
- Familiarity with high-content imaging platforms (e.g., Opera Phenix, ImageXpress, IN Cell, Yokogawa CV8000) and image analysis tools (Harmony, Columbus, CellProfiler, QuPath).

Preferred Qualifications:
- Co-culture/tri-culture/organoid neuron–glia interaction experience.
- CRISPR/Cas9 editing in human iPSCs (guide design, delivery, clone isolation, validation).
- Pooled/arrayed CRISPR screen execution (MAGeCK or comparable analysis).
- Neurodegenerative/TDP-43 (ALS/FTD) experience.
- R/Python for QC/plots and collaboration.
- AI/ML image analysis familiarity; CRISPR activation/interference.
- Pharma/biotech industry experience and GLP/GxP-adjacent documentation exposure.
- Co-authorship on peer-reviewed publications.
